What do they do/look for?
They look at the roof shingles to see how old they are and their life expectancy..
Check basement or footings for cracks
Check heating apparatus for age and efficiency.
Windows for drafts and heat loss.
Chimney for cracks or deterioration.
Brick work or siding condition.
Driveway condition.
Insulation
These are the main things checked, because they can be expensive to repair.

After a seller has accepted a buyer's offer there is the house inspection, which usually stipulates that upon finding considerable cost consuming repairs are needed the buyer may negotiate a lower selling price.
The seller may offer to repair the problems or counter offer until both buyer and seller agree on a price.
I think some of these were mentioned: quality and condition of the foundation and basement,siding, roofing, insulation,doors and windows,heating system including a lined masonary flue, electrical and plumbing systems, appliances,and general overall condition. And these days with internet access concerns the buyer may actually desire improved phone service. All these utility deficiencies may be acceptable if they are reflected in the asking price.
New homes may have more problems passing these inspections that older well maintained homes. Books are on the market detailing horror stories of shoddy workmanship by modern sleazy builders.

They also check for termites and wood worms.
They check to see if your house matches the property assessment from public records, if their are any illegally completed additions.

I would recommend before anyone BUYS a home to have not only a house inspection, but a leak detection study (About $125) to determine if there are unseen leaks. This is well worth the money, save yourself the headache.
These might not apply where you are but In the UK they also do what's called a local search - to see whether there are any plans being made which might affect your property - such as building a motorway at the end of your garden!
Also if houses are built in areas where underground mining has occured, they check for the possibility of subsidence (mining survey).
