Can't speak for liberal/mainstream faiths, but fundamentalist religion takes itself hyper-seriously (in a weird way, it kinda has to).
A sense of irony tends to undermine that sort of thing.
A sense of irony requires a certain amount of objectivity. So the wackier the belief system, the less objectivity the adherents have and therefore the less they're able to take a joke.
The lack of objectivity is enforced by the fact that a) people instinctively know that if they look at things objectively their belief system will fall apart and b) those that have a certain level of objectivity are the ones that eventually leave behind the wacky beliefs.

Agreed with everything said above. Also seems to go hand-in-hand with missing chunks of what was once known as Common Sense.
So not only can these people not laugh at themselves, they can't understand why they might need to.
And that is why laughing at them is so important. They just hate being lampooned, because it shows that those of us laughing do not take them seriously in any way.
And they desperately wish to be taken seriously.
I heard a young female comic, who I very much respect, utter the other day a just perfect line, she courageously said, in the middle of a sentence, :
" ............ The Prophet Mohammad, or as I prefer to call him, .... Mohammad......".

Phizzy-- exactly that. It has always blown my mind that the higher the religiosity, the lower the threshold for humor, joy, gentleness, authenticity. That too is why I love Eddie Izzard's send-up of institutional Christianity: God says to His son, "Jeezy Chreezy." "Don't call me Jeezy Chreezy, Dad!
