Disclaimer: freedom of subjective reaction--there is no such thing as a best riff.
I'd make a big list of personal favorites.
I like Voodoo Chile but thought of it more as a memorable blues song than a
riff song. My favorite Hendrix song is Villanova Junction.

Enter Sandman by Metallica just gets me going....
Smoke on the Water by Deep Purple has to be in the top 5 at least, right?

Far too many to pick just one. Although the first one that came to mind was Comfortably Numb.

Reeling in the years by Steely Dan is the best guitar solo riff ever.

Jimi Hendrix is great and by far one of the greatest ever but David Gilmour on Pink Floyd's Comfortably Numb's always does it for me!
Manic mama - You are person of absolute taste. IMO the best song ever written. I saw floyd three times, the orginal wall tour, Momentart lapse of reason and the 1994 tour. To see Comfortably Numb live with that enormous mirrror ball will be one the falsh backs on my deathbed.

It would seem everyone here has great taste.
Flipper I love Jimmy Page, most of my mobile ringtones are led zepp tracks.
Purple sofa great thread. Standing at the station is awesome.
Satanus, always hard to decide but I agree echoes is sublime, the initial "ping" sends shivers up my spine every time.
ATJ Metallica Rock!!!!!
Steely Dan one of my favourite bands, great to see they are featured on Guitar Hero world tour so I can introduce my kids to great music.
No one has metioned Black Sabbath / Ozzy Osbourne yet - some riffs riffs by Tomy Iomi and the gang.

Oh god, cantleave, you saw pink floyd live??!! Can i have your autograph?? Hehe. It must have been a monumental experience. I sometimes watch the pulse video. The light show alone is a spiritual experience.
