Okay
The virus responsible for Covid-19 was identified as a subspecies of Sarbecovirus, and designated as SARS-CoV-2 in late 2019

The possibility of targeting the spike protein in a vaccine was discussed from about mid 2020 onward:

None of this discussion would have been possible if the virus had not been isolated. Frankly I'm bewildered at how some people (Not necessarily you) can criticize mRNA vaccines while simultaneously claiming the virus has not been isolated. The former is just not possible without the latter.

You appear to be repeating claims from Facebook and other social media platforms.
Although I've worked in a med lab, I'm not exactly sure what you mean by "purified" vis-à-vis the virus itself. Are you perhaps referring to sample purity? Or maybe to Phenol-chloroform-RNA purification (Or something similar) as a testing method?
At any rate, the claim that the virus has never been identified can still be found on podcasts, Youtube, BitChute and other mediums that don't have to pass any scientific standard or scrutiny.
It appears to originate with a gentleman named David Crowe, a telecommunications consultant, environmentalist, and critic of science and medicine.
His claim has been repeatedly refuted:
